Stranded man survives 10 days on snow in desert

A 76-year-old diabetic Colorado man survived 10 days in the remote Nevada desert by melting snow and using skills he learned as a Boy Scout, but a friend who was with him and ventured away to get help died.

James Klemovich and Laszlo Szabo, 75, went to scope out some mines in the state when their car became stuck on a lonely road with no cell phone service, Klemovich's wife, Joanne, said Thursday.

The men tried unsuccessfully to dislodge the car, and lit flares and started fires in hopes somebody would see them in northwestern Nevada's Pershing County, an area where less than 7,000 people are spread over 6,000 square miles.

They used a towel in the car to strain ditch water and snow into water bottles, but, after four or five days, Szabo left to get help. Joanne Klemovich began to worry when several days passed without a phone call from her husband.

"I figured maybe they'd had an accident and they were stranded," she said. "I thought maybe they were in a mine shaft. All kinds of things were going through my head."

Joanne Klemovich said she was expecting the worst when authorities called Tuesday night to say her husband had been found by military personnel who were holding training exercises in the area.

"I thought it was bad news, but it was very good news," she said by telephone from the couple's home in Littleton, Colo. "I didn't know what to even do or say."

James Klemovich has diabetes, wears a pacemaker and had a triple bypass heart surgery, his wife said.

He told her he wasn't panicking while he sat for days waiting for Szabo's return, she said. He kept a journal, noting how much water he drank and what he did each day. And he wrote a letter each day for her.

Drinking regularly was likely the biggest factor in his survival despite the diabetes that could have sent his blood sugar dangerously out of whack, according to Rita Kalyani, who teaches endocrinology at the Johns Hopkins University School of Medicine in Baltimore.

During a fast, she said, the body can draw glucose from the liver or from fat stores to keep levels from dropping too low. But having enough water is essential to flush out excess glucose and prevent levels from rising too high.

When the military personnel found Klemovich, they gave him a banana, two oranges and three boiled eggs, he told his wife.

Szabo, of Lovelock, Nev., was found dead about a mile and a half away. An autopsy is being performed.

Klemovich said her husband hasn't been talking much about his friend and that she doesn't know whether Szabo has any close relatives.

"When I first talked to him I could tell he'd been through an ordeal," she said. "When he called back, he sounded pretty good."

James Klemovich is still in Nevada, waiting for the car to be recovered before returning home. He was treated and released from a hospital.

Joanne Klemovich said she knows the first words she'll say to her husband of 48 years as of Wednesday: "Happy belated anniversary."

A new chemical analysis of lunar material collected by Apollo astronauts in the 1970s conflicts with the widely held theory that a giant collision between Earth and a Mars-sized object gave birth to the moon 4.5 billion years ago.

In the giant-collision scenario, computer simulations suggest that the moon had two parents: Earth and a hypothetical planetary body that scientists call "Theia." But a comparative analysis of titanium from the moon, Earth and meteorites, published by Junjun Zhang, graduate student in geophysical sciences at the University of Chicago, and four co-authors indicates the moon's material came from Earth alone.

If two objects had given rise to the moon, "Just like in humans, the moon would have inherited some of the material from the Earth and some of the material from the impactor, approximately half and half," said Nicolas Dauphas, associate professor in geophysical sciences at UChicago, and co-author of the study, which appears in the March 25 edition of Nature Geoscience.

"What we found is that the child does not look any different compared to the Earth," Dauphas said. "It's a child with only one parent, as far as we can tell."

The research team based their analysis on titanium isotopes ? forms of titanium that contain only slight subatomic variations. The researchers selected titanium for their study because the element is highly refractory. This means that titanium tends to remain in a solid or molten state rather than becoming a gas when exposed to tremendous heat. The resistance of titanium isotopes to vaporization makes it less likely that they would become incorporated by the Earth and the developing moon in equal amounts.

Titanium also contains different isotopic signatures forged in countless stellar explosions that occurred before the sun's birth. These explosions flung subtly different titanium isotopes into interstellar space. Different objects in the newly forming solar system gobbled up those isotopes in different ways through collisions, leaving clues that let scientists infer where the solar materials including the moon came from.

Planetary DNA

"When we look at different bodies, different asteroids, there are different isotopic signatures. It's like their different DNAs," Dauphas said. Meteorites, which are pieces of asteroids that have fallen to Earth, contain large variations in titanium isotopes. Measurements of terrestrial and lunar samples show that "the moon has a strictly identical isotopic composition to the Earth," he said.

"We thought that the moon had two parents, but when we look at the composition of the moon, it looks like it has only one parent," Zhang said.

Zhang initially found variations in the titanium isotopic composition between the lunar and terrestrial samples. She then corrected the results for the effects of cosmic rays, which could have changed the titanium isotopic composition of the lunar samples.

The Earth and the moon are constantly bombarded by cosmic rays from the sun and from more distant sources in the galaxy. Earth's atmosphere and magnetic field prevents most of these rays from reaching its surface, but the moon has no such protection.

"We compared the titanium isotopic composition with samarium and gadolinium since those two systems are very sensitive to the cosmic-ray effect," Zhang said. The only compositional differences the scientists expected to see in samarium and gandolinium between Earth and moon would be the result of cosmic rays. "We found a very nice linear correlation between titanium and samarium or gadolinium," she said.

Zhang's titanium analyses greatly reinforce previous work by other researchers who came to the same conclusion after comparing terrestrial and lunar oxygen isotopes, which are less refractory and thus more likely to gasify during a giant impact than titanium.

Lunar Conundrum

Solving the conundrum of the moon's origin probably will prove challenging because all of the alternative scenarios for the moon's formation have drawbacks.

For example, it is possible that even though titanium is refractory, it might still have gasified in the giant impact and then became incorporated into the disk of Earth-orbiting material that developed into the moon. This might have erased the signature of the titanium from Theia, which could explain the UChicago team's observations. The problem with this scenario is that the disk may have fallen back to Earth if too much material was exchanged between the two bodies.

An old idea, long abandoned, is that the moon arose via fission from a molten, rapidly rotating Earth following a giant impact. This idea explains the similarity between Earth and moon, but how such a large, concentrated mass could spin fast enough to split in two remains problematical.

According to a third scenario, Earth collided with an icy body lacking entirely in titanium. There are no bodies made purely of ice in the solar system, however. "They would always have a significant fraction of solid material, so you would still expect the object to deliver some titanium," Dauphas said.

It's also possible that Theia had the same composition as Earth. This is unlikely, however, because of the widely accepted view that the Earth incorporated material over tens of millions of years in collisions with smaller bodies that flew in from different regions of the developing solar system.

"We thought we knew what the moon was made of and how it formed, but even 40 years after Apollo, there is still a lot of science to do with those samples that are in curatorial facilities at NASA," Dauphas said.

Plant research reveals new role for gene silencing protein

ScienceDaily (Mar. 29, 2012) ? A DICER protein, known to produce tiny RNAs in cells, also helps complete an important step in gene expression, according to research on Arabidopsis thaliana. The expression of a gene, when an organism's DNA is transcribed into a useable product, requires activation via a promoter or an external trigger. Plant research to be published in Science helps to show that later stages of transcription are just as important. This is likely to apply to other organisms, including humans.

Termination is the final stage of transcription. Successful termination is dependent on DNA being transcribed into RNA with the correct sections, including a certain length tail.

Scientists at the John Innes Centre on Norwich Research Park have found that where effective termination through the normal mechanisms has not occurred, DICER-LIKE 4 (DCL4) steps in to tidy up. Without termination, transcription continues down the chromosome unchecked.

In this way, DCL4 plays a crucial and previously unknown role in transcription termination. It helps formation of the gene product. DCL4 is more commonly known to play a part in the opposite effect, gene silencing.

"DCL4 is a back-up to termination processes, helping a gene to be successfully expressed," said lead author Professor Caroline Dean from JIC, which is strategically funded by BBSRC.

The findings may help explain why gene silencing happens so often with transgenes. It was not known that so much attention should be given to the tail end of a gene.

"Our research shows that for successful expression the end of a gene is just as important as its beginning," said Dean.

When termination fails a lot of aberrant RNA is made -- this is degraded as part of a cell's quality control mechanism. This can have consequences for other sequences in the genome that match the aberrant RNA.

"If a gene ends badly, aberrant RNA will trigger silencing pathways," said Dean.

DCL4's ability to step in to rescue poor termination makes it important for both successful gene expression, a previously unknown role for it, and gene silencing.

The research was funded by JIC's strategic funding from BBSRC and by the EU research project SIROCCO, focused on silencing RNAs

Discovery provides blueprint for new drugs that can inhibit hepatitis C virus

ScienceDaily (Mar. 19, 2012) ? Chemists at the University of California, San Diego have produced the first high resolution structure of a molecule that when attached to the genetic material of the hepatitis C virus prevents it from reproducing.

Hepatitis C is a chronic infectious disease that affects some 170 million people worldwide and causes chronic liver disease and liver cancer. According to the Centers for Disease Control and Prevention, hepatitis C now kills more Americans each year than HIV.

The structure of the molecule, which was published in a paper in this week's early online edition of the journal Proceedings of the National Academy of Sciences, provides a detailed blueprint for the design of drugs that can inhibit the replication of the hepatitis C virus, which proliferates by hijacking the cellular machinery in humans to manufacture duplicate viral particles.

Finding a way to stop that process could effectively treat viral infections of hepatitis C, for which no vaccine is currently available. But until now scientists have identified few inhibiting compounds that directly act on the virus's ribonucleic acid (RNA) genome -- the organism's full complement of genetic material.

"This lack of detailed information on how inhibitors lock onto the viral genome target has hampered the development of better drugs," said Thomas Hermann, an associate professor of chemistry and biochemistry at UC San Diego who headed the research team, which also included scientists from San Diego State University. The team detailed the structure of a molecule that induces the viral RNA to open up a portion of its hinge-like structure and encapsulate the inhibitor like a perfectly fit glove, blocking the ability of the hepatitis C virus to replicate.

The molecule is from a class of compounds called benzimidazoles, known to stop the production of viral proteins in infected human cells. Its three-dimensional atomic structure was determined by X-ray crystallography, a method of mapping the arrangement of atoms within a crystal, in which a beam of X-rays strikes a crystal and causes the beam of light to spread. The angles and intensities of the light beams allowed the scientists to calculate the structure of the viral RNA-inhibitor complex.

"This structure will guide approaches to rationally design better drug candidates and improve the known benzimidazole inhibitors," said Hermann. "Also, the crystal structure demonstrates that the binding pocket for the inhibitors in the hepatitis C virus RNA resembles drug-binding pockets in proteins. This is important to help overcome the notion that RNA targets are so unlike traditional protein targets that drug discovery approaches with small molecule inhibitors are difficult to achieve for RNA."

Mukherjee puts money in one pocket, takes out from others (Roundup)

New Delhi, March 16

Every tax payer in India is set to gain at least Rs.2,000 per annum, thanks to the relief proposed by Finance Minister Pranab Mukherjee. But the happy story ends there! With a hike of 2 percentage points in service tax and higher excise and customs duty on a host of other items, your household budget is actually set to rise.

In his seventh national budget presented in the Lok Sabha Friday, Mukherjee raised the income tax exemption limit for individuals to Rs.2,00,000 from Rs.1,80,000 and revised the tax slabs to bring relief to some 35 million people in the country.

But by bringing virtually all services under the tax net and by hiking the rate from 10 percent to 12 percent -- as also by raising the standard excise rate to 12 percent from 10 percent -- he also ensured you pay more indirect taxes than before.

This is why the net impact of his proposals is such that while direct tax measures will result in a net revenue loss of Rs.4,500 crore ($900 million), the indirect tax changes will fetch a net revenue gain of Rs.45,940 crore ($9.1 billion) -- over 10 times more.

You may have to pay more for dining out, gold jewellery, luxury cars and smoking, even as you get relief while watching a movie in cinema halls, investing in equities, going for a preventive health check or paying for your child's education.

On income tax front, the finance minister proposed new tax slabs in which income up to Rs.2,00,000 will be totally exempt, levy 10 percent on Rs.2,00,000 to Rs.5,00,000, then 20 percent on Rs.5,00,000 to Rs.10,00,000 and 30 percent on income above Rs.10,00,000.

"This will provide tax relief of Rs.2,000 to every tax payer," the finance minister said, adding: "My proposal on direct taxes will result in a revenue loss of Rs.4,500 crore."

Overall, in the Rs.14,90,925 crore ($300 billion) budget, the fiscal deficit -- which is the total projected expenditure less tax and non-tax revenue, as also loan recoveries -- is pegged at 5.1 percent of GDP, which is lower than 5.9 percent for the current fiscal.

He said with the economy getting back on rails, it was time for some tough measures -- a statement endorsed by Prime Minister Manmohan Singh who said it was necessary to "bite the bullet", especially on issues such as subsidies.

"When the time comes for taking tough decisions, we hope we will be able to take all our allies on board," the prime minister said, giving his reactions to the budget when asked specifically as to how he hoped to carry the coalition partners along.

Even Mukherjee said it was more important to address the challenges than earn brownie points. "Whether or not today's announcements make tomorrow's headlines matters little, as long as they help in shaping the headlines that describe India a decade from now.

For the corporate sector, the finance minister, said, while the tax rates will remain unchanged, he assured cheaper access to funds for expansion, even as he tinkered with the excise rates and customs duties for specific items.

Mukherjee said he also proposed to tax every service, but for 17 items specified in what he termed as a negative list, and enhanced the rate of such levy to 12 percent from 10 percent.

This is projected to fetch Rs.1,24,000 crore ($24.8 billion) against revised estimate of Rs.95,000 crore ($19 billion) for 2011-12 .

Assuring further liberalisation of capital markets, he announced a new equity savings scheme to extend income tax deduction of 50 percent to those who invest up to Rs.50,000 in equities and whose annual income is less than Rs.10 lakh.

Mukherjee spelt out some far-reaching reforms in the budget, notably on the subsidy front, which he promised to cut to 2 percent of India's gross domestic product (GDP).

"Such a step is needed to improve the quality of public spending," he said, adding the effort now will be to directly transfer subsidies on fertiliser, food and fuels to to the intended beneficiaries.

He hoped for an early implementation of the direct tax code and a pan-India goods and services tax to unify federal and state taxes, even as he promised incentives to attract capital for infrastructure development.

He also said the government will make an all-out effort to curb outflow of black money and bring back such ill-gotten funds stashed away in tax havens abroad.

Mukherjee began his speech touching on the cascading ill-effects of the global slowdown on the Indian economy, but assured people there were clear signs of recovery that should see the country grow at 7.6 percent in 2012-12, against 6.9 percent this fiscal.

"The global crisis has affected us. India's gross domestic product (GDP) is expected to grow at 6.9 percent in 2011-12, after having grown at 8.4 percent in each of the two preceding years," he said.

"Though we have been able to limit the adverse impact of the slowdown in our economy, this year's performance has been disappointing. But it is also a fact that in any cross-country comparison, India still remains among the front-runners in economic growth."

At the same time, Mukherjee also said the Indian economy was at the "cusp of a revival", as agriculture and services have continued to grow at a decent pace. It was industrial performance that was acting as a drag.

"While we do not have the aggregate figures for the last quarter of 2011-12, numerous indicators pertaining to this period suggest that the economy is now turning around. There are signs of recovery in coal, fertiliser, cement and electricity sectors."

(AP) ? The public radio program "This American Life" on Friday retracted a story about what a monologist said he found while investigating Apple operations in China, citing "numerous fabrications."

The show's Friday broadcast will detail inconsistencies in the highly popular Jan. 6 episode that was an excerpt from writer Mike Daisey's critically acclaimed one-man show "The Agony and the Ecstasy of Steve Jobs," which currently is at the Public Theater in New York.

"We're retracting the story because we can't vouch for its truth," Ira Glass, host of "This American Life," said in a letter posted on the show's website. Spokeswoman Emily Condon said Glass wouldn't take calls for comment until after Friday's episode airs.

In his program, Daisey describes meeting workers who put in very long hours and were forced to do crippling, repetitive motions at factories that make Apple products in China.

But "This American Life" says Rob Schmitz, a China correspondent for the public radio show "Marketplace," located and interviewed Daisey's Chinese interpreter, who disputed much of the artist's claims.

"This American Life" said in its statement that staffers asked Daisey for his interpreter's contact information while fact-checking the story and he said the cellphone number he had for her didn't work anymore and he had no way to reach her.

"At that point, we should've killed the story," Glass said in the statement. "But other things Daisey told us about Apple's operations in China checked out, and we saw no reason to doubt him."

Daisey posted on his web site Friday that he stands by his work and that what he does is theater, not journalism.

"'This American Life' is essentially a journalistic ? not a theatrical ? enterprise, and as such it operates under a different set of rules and expectations. But this is my only regret," Daisey's letter said.

Apple has been rebutting Daisey's allegations for months, to little effect. An Apple spokeswoman declined comment Friday.

The original episode, "Mr. Daisey and the Apple Factory," has become the most popular podcast in the history of "This American Life" with nearly 890,000 downloads.

In his show, Daisey splices career milestones of Steve Jobs and the transformation of Apple from a David into a Goliath with more personal stories about his own connection to the computer maker.

He has said that when he saw four photos posted online taken by workers at a Chinese factory to test the iPhone but mistakenly not erased, he suddenly realized people, not robots, were putting the sleek devices together.

In interviews and on stage, Daisey has said he traveled to the Chinese industrial zone of Shenzhen and interviewed hundreds of workers from Foxconn Technology Group, the world's largest electronics contract manufacturer.

"I talk to people whose joints in their hands have disintegrated from working on the line, doing the same motion hundreds and hundreds of thousands of times. It's like carpal tunnel on a scale we can scarcely imagine," he said while performing the show in New York in October.

While the piece specifically targets Apple, most of what Daisey said he discovered is applicable to many high-tech manufacturers. Daisey has performed the new monologue for some 50,000 people from Seattle to Washington, D.C., and it is now at The Public Theater until Sunday. Daisey was expected to take the show on tour, but its future is now in doubt.

In a statement, The Public Theater said the show would be performed in New York as scheduled and stood by what it called "a powerful work of art."

"Mike is an artist, not a journalist," the statement said. "Nevertheless, we wish he had been more precise with us and our audiences about what was and wasn't his personal experience in the piece."

In preclinical studies, researchers at SRI International and Astraea Therapeutics have recently evaluated the role of a new drug receptor target that shows promise for the treatment of drug addiction.

This potential new drug target belongs to a class of receptors called the nicotinic acetylcholine receptors (nAChRs). One subtype of nAChRs, called alpha4beta2 is a well-known target for nicotine's addictive effects and the therapeutic effect of the smoking cessation drug varenicline. SRI researchers are now studying another, lesser-known subtype, called alpha3beta4 nAChR, which has recently shown to play a role in the addictive properties of cocaine, morphine, and nicotine.

Using AT-1001, a highly selective alpha3beta4 compound developed by Astraea Therapeutics, SRI researchers found that addiction processes could be disrupted. In preliminary studies, AT-1001 also reduced nicotine withdrawal symptoms such as anxiety.

Because addictive drugs act on the brain's "reward circuit," many people compulsively take them despite their harmful consequences. Feelings of pleasure are caused by the release of brain chemicals, or neurotransmitters, and their increased activity in the brain reward circuit. The key neurotransmitter in the reward circuit is dopamine, which produces changes in addictive behavior. Researchers speculate that disruption of the alpha3beta4 nAChR system using highly selective drugs such as AT-1001 may interrupt this reward circuitry.

"Currently, there are no therapeutics approved to treat addiction to stimulants such as cocaine, so it is intriguing to find a promising drug receptor target and to see that AT-1001 can indeed affect phenomena that are thought to be deeply tied to the addictive nature of cocaine and other drugs of abuse," said Taline Khroyan, Ph.D., Senior Behavioral Pharmacologist in SRI's Center for Health Sciences.

Findings from these collaborative studies were recently presented at the Society for Neuroscience Annual Meeting in a study titled "AT-1001, a Highly Selective alpha3beta4 nicotinic Receptor Antagonist is Effective in Blocking Place Preference, Behavioral Sensitization, and Withdrawal Effects of Various Drugs of Abuse." The presentation was highlighted by the Society for describing the innovative approach taken by the research team.

Those were the opening remarks of a segment on Tuesday evening's episode of the Glenn Beck Program in which Glenn revealed how he was threatened by none other than rabid Antisemite Louis Farrakhan -- or at least by his henchmen at The Nation.

The NOI official newspaper wrote over the weekend that?Glenn Beck ?is a minion of the Synagogue of Satan?and as such represents the thinking of those used as their low-level pawns.?

The author continued: ?The Holy Qur?an teaches Muslims to never be the aggressor in word or deed, but if attacked, we fight with those who fight with us.?

Sounds a tad threatening, no?

Glenn responded on his program by taking NOI and Farrakhan to task, exploring the movement and their vitriolic leader's history of violence. He also delved into the reasons why he said Farrakhan and NOI might be a threat to President Obama.

Many will recall the fate that befell Malcolm X once he broke with The Nation.

COMMENTARY | As a combat veteran, I can attest that everyone has their breaking point. The moment when all normal mental functions cease and you do something no sane person would. A temporary lapse in proper judgment has been the fall of a great many good men, and it is sad that it apparently has happened again.

According to MSNBC, an American soldier is accused of killing 16 civilians in Afghanistan. The yet to be named soldier was on his first deployment to the country. It is also reported that since 2003, his brigade had been to Iraq three times.

Obviously not condoning the horrific actions, but if you want a reason as to why a man would snap, there it is. As much as the U.S. military likes to tell us it is doing as much as possible to combat the symptoms of post traumatic stress disorder, it is forgetting the No. 1 trigger: Sending men and women back repeatedly to the place that causes them the stress. Nobody should be sent back to a combat zone as many times as this individual's brigade is said to have been.

But the horrific killing of 16 people has happened. You know people are going to line up around the block looking for an excuse to go after the U.S. According to CNN, the Taliban is vowing revenge. Not to be flip, but the Taliban wants revenge for everything, from a murder to the U.S. letting women drive cars. The Taliban needs to realize its stock isn't quite what it once was. A call of revenge from the Taliban is like a 5-year-old saying he's going to beat up Mike Tyson.

According to BBC News, an Afghan intelligence official said this was a propaganda victory for the Taliban. Again, proof the terrorist group will use anything to further its agenda. There is too much international pressure from all sides on the group. It does nobody any good to use a killing to its advantage.

If the Afghans want to take this individual and try him in their courts, I see no fault in it. But U.S. officials should realize that something like this could have been prevented if it just didn't send the same people back so many times.
